Aripiprazole

Indications/Uses

Schizophrenia.

Dosage/Direction for Use

IM inj Starting & maintenance dose: 400 mg mthly (no sooner than 26 days after the previous inj). May reduce dose to 300 mg once mthly if there are AR.

Contraindications

Hypersensitivity.

Special Precautions

Increased mortality in elderly patients w/ dementia-related psychosis; cerebrovascular AR, eg, stroke in elderly patients w/ dementia-related psychosis; NMS; tardive dyskinesia; metabolic changes; pathological gambling & other compulsive behaviors; orthostatic hypotension; leukopenia, neutropenia & agranulocytosis; seizures; potential for cognitive & motor impairment, body temp regulation; dysphagia, QT interval, falls, venous thromboembolism, priapism, akathisia.

Adverse Reactions

Increased wt, akathisia, inj site pain, sedation. Abdominal discomfort, constipation, diarrhoea, dry mouth, toothache, vomiting.

Drug Interactions

Increased exposure w/ strong CYP3A4 inhibitors (eg, ketoconazole) or CYP2D6 inhibitors (eg, paroxetine, fluoxetine). Decreased exposure w/ strong CYP3A4 inducers (eg, carbamazepine). May enhance effect of certain antihypertensive agents. Increased intensity of sedation & orthostatic hypotension in combination w/ benzodiazepines (eg, lorazepam).
