Indications and Dosage
Ophthalmic
Superficial ocular infections Adult: As oint containing bacitracin 500 U and polymyxin B 10,000 U/g: Apply to affected eye(s) 3-4 hrly for 7-10 days, depending on severity of infection. Topical/Cutaneous Superficial infections Adult: As oint containing bacitracin 500 U and polymyxin B 10,000 U/g: Apply thinly to affected area 1-3 times daily, may be covered w/ a sterile bandage.
Child: Same as adult dose. |

Contraindications
Hypersensitivity to bacitracin or polymyxin B.

Special Precautions
Prolonged use may cause overgrowth of nonsusceptible organisms, including fungi. Pregnancy and lactation.

Adverse Reactions
Burning sensation, local irritation, swelling, conjunctival erythema, pruritus (ophth). Rarely, anaphylaxis.

Action
Description:
Mechanism of Action: Bacitracin interferes w/ bacterial cell wall synthesis by inhibition of the regeneration of phospholipid receptors involved in peptidoglycan synthesis. It is bactericidal to a number of gram +ve and –ve organisms. Polymyxin B increases the permeability of the bacterial cytoplasmic membrane by interacting w/ the phospholipid components. It is bactericidal to a number of gram –ve organisms. Pharmacokinetics: Absorption: Poorly absorbed from intact or denuded skin, wounds, or mucous membranes. |
