Hydroxychloroquine sulfate
Indications/Uses
SLE.
Dosage/Direction for Use
Adult Max: 6.5 mg/kg daily based on ideal body wt 1-2 times daily. Patient who received >400 mg Initial dose: 400 mg in divided doses. Decrease dose into 200 mg if there is no significant changes. Maintenance dose may be increased to 400 mg if needed. Childn >31 kg Do not exceed 6.5 mg/kg daily based on ideal body wt.
Administration
Should be taken with food.
Contraindications
Hypersensitivity to 4-aminoquinolone compd. Eye maculopathy. Childn <6 yr (<31 kg).
Special Precautions
Eye exam related to the risk of retinopathy before & after treatment. Low visual acuity. Hypoglycemia. Pregnancy & lactation. Elderly.
Adverse Reactions
Retinopathy, cardiomyopathy, Torsades de pointes allergic reaction, skin rash, pruritus, urticaria, bronchospasm, angioderm, nausea, diarrhea, alopecia, anorexia, skeletal muscle myopathy or neuropathy.
Drug Interactions
May increase serum levels of digoxin. Reduction of insulin dose or antidiabetic drugs may be needed. May prolong QT interval & increase risk of ventricular arrhythmias w/ drugs that have the potential to induce cardiac arrhythmias eg, metfloquine. Antiepileptic drug activity may be disrupted. May increase side effects w/ MTX. Increased levels of cyclosporine.
